Thompson attended Pattonville High School and was the drummer in the school band. She graduated from Pattonville in 1999 having earned a full scholarship to the Manhattan School of Music, which she graduated from in 2003 with a bachelor's degree in Jazz Composition and Education.

Thompson founded KTMUSIC Productions, a record label based in Harlem, New York. In 2010, she released her first album via the label, Like Clockwork. Thompson has served as session or touring drummer with several artists, including Raymond Angry, John Blackwell, Richard Bona, Terri Lyne Carrington, Dennis Chambers, Gary Dourdan, Roy Hargrove, Anthony Jackson, Jay-Z, Jason Moran, Meshell Ndegeocello, Reverend Run, Wallace Roney and Mike Stern. She was also a longtime drummer for Beyoncé, having begun touring with her in 2006.
In addition to performing with other artists, Thompson has performed with variations of her own band. In 2009, she performed with The Kimberly Thompson Quartet, which featured band members Mark Shim (saxophone), Craig Magnano (guitar), and Tarus Mateen (bass). In 2012, Thompson performed with The Kimberly Thompson Quintet, which featured the lineup of Eden Ladin (piano), Philip Dizack (trumpet), Dayna Stephens (tenor sax), and Ben Meigners (bass).
