Blackmore said the first attack occurred at 5:40 a.m. CST, with subsequent emergency calls reporting more stabbings.

The two suspects wanted in connection to the stabbings were identified as Damien Sanderson, 31, and Myles-Brandon Sanderson, 30. They were believed to be driving a black Nissan Rogue and were reportedly seen in Melfort and the Arcola Avenue area of Regina. They are "armed and dangerous" according to the RCMP. It is currently unknown where the suspects may be going, or if they changed vehicles since the last known sighting of them.

STARS medical helicopters and ambulances were dispatched to triage and transport wounded victims to hospitals. Fifteen injured victims were reported, but there may others who took themselves to hospitals.
In response to the stabbings, the Joints Chiefs and Councils of the James Smith Cree Nation declared a local state of emergency at 12:00 p.m. CST on the day of the stabbing. The state of emergency will be effective until 5:00 p.m. CST on September 30. Additionally, the community set up two emergency operation centers.
At 7:12 a.m. CST, the RCMP issued an emergency alert within the region, advising residents to stay in secure locations and to use caution when allowing others into their homes. It also alerted citizens not to approach the suspects and instead report them to 9-1-1. At 8:20 a.m. CST, the alert was re-issued province-wide and with information on the suspect's vehicle. At the request of the Saskatchewan RCMP, the alert was later expanded to include all of Alberta and Manitoba.
The Saskatchewan Health Authority (SHA) initiated a code orange, issuing additional staff in hospitals local to the stabbings to help with the influx of patients. This was later declared over, as the influx of new patients had subsided.
The attacks occurred on the same day as the Saskatchewan Roughriders' annual Canadian Football League (CFL) rivalry game against the Winnipeg Blue Bombers; team officials stated that there would be an enhanced police presence at Mosaic Stadium.
Reactions
Premier Scott Moe stated that "there are no words to adequately describe the pain and loss caused by this senseless violence." Prime Minister Justin Trudeau called the stabbings "horrific and heartbreaking", and stated that "I'm thinking of those who have lost a loved one and of those who were injured".
