The Women's 4 × 400 metres relay at the 2010 Commonwealth Games as part of the athletics programme was held at the Jawaharlal Nehru Stadium on Monday 11 October and Tuesday 12 October 2010.

On 15 October 2010, three days following the event, Folashade Abugan was reported to have tested positive for Testosterone prohormone, subsequently disqualifying her results including those of the Nigerian women's 4 x 400 metres relay team. The English and Canadian teams' results were elevated to the silver and bronze medal-winning positions respectively.
